FIELD: biotechnology, biochemistry, enzymes. SUBSTANCE: strain-producer is isolated from colonial ascidium inhabiting near island Shikotan (Kurile Isls) and deposited in collection of Marine Microorganisms TI BOCH DVO RAN at = 4628. Method for preparing endonuclease involves culturing the strain-producer in nutrient medium, extraction of culture with 2.5 volumes of 0.03 M Na-acetate buffer, pH 3.2), heat treatment of extract at 75 C, separation of solid part and the following fractionating and concentrating proteins on chitosan sorbent. Purification of crude enzyme preparation is carried out using gel-chromatography on Sephadex G-75. Invention relates to preparing enzymes and can be used for preparing highly active endonuclease Pest [EC 3.1.4.21] that represents a single- -strand nucleate-5'-oligonucleotide hydrolase. The latter can be used in molecular biology in investigations of primary structure of deoxyribonucleic acid and structure of ribosomal, transport and viral ribonucleic acid, in genetic engineering for preparing recombinant molecules and for preparing 5'-ribo- and 5'-deoxyribonucleotides from ribonucleic acid and deoxyribonucleic acid.
