METHOD FOR VISUALIZATION OF POULTRY FORM BLOOD ELEMENTS UPON SINGLE SMEAR Russian patent published in 2004 - IPC

FIELD: veterinary science. SUBSTANCE: sample of unstabilized blood is supravitally dyed with brilliant cresyl blue dyestuff for 20-40 min at 18-20 C. Moreover, premixing of blood with dyestuff should be conducted laminally in closed volume for 30 sec at the same temperature. After preparing and drying the smears one should conduct fixation in fixing solution by Leischman for 3 min to further finish dyeing with diluted buffer solution at pH = 6.8-7.2 consisting of 0.95% disubstituted sodium phosphate and 0.907% monopotassium phosphate as fixing dyestuff by Leischman for 10 min. Smear should be washed with the same buffer solution. The suggested method enables to visualize erythrocytes, reticulocytes, basophiles, eosinophils, pseudoeosinophiles, lymphocytes, monocytes and thrombocytes simultaneously upon single smear.
