FIELD: preparative biochemistry, enzymology. SUBSTANCE: invention describes a method for preparing affine sorbent used for purification of enzyme preparations that involves interaction amino-containing insoluble carrier with peptide ligand and condensing agent in an organic solvent medium. As a peptide ligand method involves using polypeptide antibiotic polymyxin M or a modified polypeptide mimicking structural motifs of collagen molecule chains and have the sequence: Z-Gly-DL-Pro-Gly-, Pro-Gly-Gly- or (Pro-OH)-Gly-Gly-. Process is carried out at pH 5.5-8.0 taking 9-80 mg of peptide ligand and 10-500 mg of condensing agent per 1 g of carrier. Invention provides the possibility for isolation of highly purified and highly active enzyme preparations used in medicine, biochemistry, food industry and in enzymatic synthesis of peptides, and for investigations aims also.
