FIELD: medicinal immunology.
SUBSTANCE: invention relates to methods for determination of functional activity of complement factors in human serum blood and can be used in diagnosis of some diseases and in biological preparations. Method involves sorption of nonspecific immunoglobulin G, addition of guinea pig serum-containing solution and sodium ethylenediaminetetraacetate. After incubation and plate drying analyzed sample containing C1r2s2 complex of human complement with unknown activity, human serum blood pseudoglobulin fraction, and buffer solution containing calcium and magnesium are added. Incubation is carried out and after a plate washing out and drying conjugate of enzyme with antibodies raised against human component C4 and substrate for this enzyme are added. Then activity of C1r2s2 complex is calculated by amount of formed product of enzymatic reaction. Invention provides the development of method allowing carrying out assay of functional activity of C1rs2 complex of human complement.
