FIELD: biology, genetic engineering, medicine, endocrinology, neuroendocrinology.
SUBSTANCE: invention proposes a method for creature of transgenic mouse no comprising functional receptor-1 for corticotropin releasing-factor. Proposed method involves preparing positive embryonic stem cells by insertion to them of transgene of recpetor-1 for corticotropin releasing-factor. Then prepared positive embryonic stem cells are inserted into murine blastocyst with the following development transgenic mouse from its. Except for, invention proposes a method for identifying agonist of releasing-factor for corticotropin, urocortin or ligand of family of corticotropin releasing-factor. This method involves insertion of testing compound or placebo compound of transgenic mouse, parallel administration of a releasing-factor for corticotropin, urocortin or ligan of family of corticotropin releasing-factor from mouse of wild type and the following evaluation and comparing the level of anxiety response, endocrine response for stress and rhythms of locomotor activity in transgenic mice and mice of wild type. Except for, invention proposes a method for identifying antagonist of a releasing-factor for corticotropin, urocortin or ligand of family of corticotropin releasing-factor. This method involves administration of testing compound or placebo compound in transgenic mouse, parallel administration of a releasing-factor for corticotropin, urocortin or ligand of family of corticotropin releasing-factor of mouse of wild type, the following evaluation and comparing the level of anxiety response, endocrine response for stress and rhythms of locomotor activity in each mouse. Also, invention proposes a screening method of compounds that are analogs or agonists of corticosterone or corticotropin using such mouse. This method involves administration of testing compound in transgenic mouse female after conception followed by estimation of lungs state in progeny obtained from this female. The proposed transgenic mouse can be used in endocrinology and neuroendocrinology as a model animal for analysis of the role of a releasing-factor receptor in the development of organism and evaluation of its biological functions. Proposed methods can be used for preparing antagonists of receptor-1 for corticotropin a releasing-factor that can be used in treatment or diagnosis of different neuropsychiatric disturbances and for the improvement of teaching ability and memory in cases associated with feeble-mindedness.
EFFECT: improved preparing method, valuable biological and medicinal properties of transgenic mouse.
