FIELD: gene engineering.
SUBSTANCE: it is necessary to isolate genomic clone containing locus of CRFR2 gene. Then comes constructing a guide vector where exons 10,11 and 12 of CRFR2 are replaced with a cassette of selective marker gene, and its transfer into embryonic stem cells. Then comes injecting embryonic stem cells into blastocysts to be then transferred for pseudopregnant female mice. Finally, one should obtain homozygous according to destroyed allele CRFR2 gene. Moreover, it is suggested to apply the method for screening compounds that control blood pressure via the way of receptor 2 corticoliberin by applying the obtained transgenic mice. The innovation enables to obtain transgenic mice which do not express protein of receptor 2 corticoliberin in no allele. The innovation could be applied in the field of neurobiology, endocrinology and psychiatry to study anxiety and depression and, also, to obtain preparations for treating these diseases.
EFFECT: higher efficiency.
