FIELD: genetic engineering, molecular biology, biochemistry.

SUBSTANCE: invention can be used in the development and realization of many methods used for DNA analysis. Invention proposes modified forms of thermostable DNA polymerases obtaining by replacing glutamate residue at position 4 of 11-membered critical motif belonging to native form of enzyme in microorganism P. thermus with other amino acid residue that results to reducing level of discriminating ability of enzyme with respect to nucleotides labeled with dye of fluorescein class. The preferable change is replacement of glutamate in indicated position with lysine. Invention describes DNA sequences encoding proposed variants of thermostable DNA polymerase, vectors for expression of these sequences in different types of host-cells, method for preparing recombinant forms of modified DNA polymerase and sets for realization of analysis with their using that simplifies the experimental procedure significantly.

EFFECT: improved preparing method, valuable biological properties of DNA polymerase.
