FIELD: medicine.
SUBSTANCE: method involves producing conditioned medium and culturing fragments. Human embryo bone tissue fragments are used for producing the conditioned medium. Bone fragments are cultured during 8-10 weeks. The medium is completely changed every week and then protein fraction, possessing fibroblast- and fibrosis-inhibiting activity, is separated by means of exchange chromatography, gel filtration and centrifuging on membrane having pore diameter of 10-100 kD. The growth factor of protein nature being purified protein is obtained with molecular weight of approximately 100 kD, soluble in water, thermostable, having fibroblast-inhibiting activity which degree is within the limits of 40% to 0% and manifesting inhibiting effect when the line grows in culturing medium containing 30% of conditioned embryo bone tissue. The protein nature growth factor is cleaned specific protein obtained or separated in a medium conditioned with embryo bone tissue with molecular weight of approximately 100 kD, soluble in water, thermostable, having fibroblast-inhibiting activity which degree is within the limits of 40% to 0% and manifesting inhibiting effect when the line grows in culturing medium containing 30% of conditioned embryo bone tissue. The protein nature growth factor is used as new fibroblast proliferation inhibitor.
EFFECT: direct anti-fibrotic activity; no action upon normal cell proliferation being observed.
