FIELD: animal science, genotyping in cattle.
SUBSTANCE: the present innovation deals with genotyping in cattle by the alleles of BoLA DRB 3 gene of the main histocompatibility complex (MHC) class II to detect animals predisposed to persistent lymphocytosis. Exon2 of BoLA DRB 3 gene should be amplified, (the size of PCR-product corresponds to 284 pn) followed by the analysis of restriction polymorphism of this area due to endonucleases Rsa I, Hae III, BstYI. If at restriction by Rsa I it is possible to detect the fragments 141, 54-51, 39 pn, amplificate should be similarly treated with restrictase Bst 2U I. In this case, allelic variant BoLA DRB 3*7 forms the fragments 24, 57, 89 and 111 pn; allelic variant BoLA DRB 3*8 - 24, 80 and 180 pn; allelic variant BoLA DRB 3*9 - 24, 57 and 203 pn. Thus, it is possible to successfully differentiate alleles BoLA DRB 3*7, BoLA DRB 3*8 and BoLA DRB 3*9. The innovation enables to detect leucosis-sensitive animals at allel BoLA DRB 3*8.
EFFECT: higher accuracy of detection.
