FIELD: biotechnology.

SUBSTANCE: molecular genetics in agriculture, used to determine the genetic resistance of cattle to the leukemia virus by identifying the *0902 allele, associated with resistance to leukemia, and the *1501 allele, associated with susceptibility to this disease, by polymerase chain reaction (PCR) with primers limiting a fragment of 216 nucleotides in length within the second exon of the bovine BOLA-DRB3 gene, followed by sequencing and computer recognition using reference sequences. The proposed method includes the BDRB32ex software script, created for computer automatic recognition of alleles of the BOLA-DRB3 gene using reference sequences. For PCR, primers ex2_for - 5 are used' -GTGTCATTTCTTCAACGGGAC-3' and ex2_rev - 5' -CGACCCCGTAGTTGTGTCTG-3'. The product of the sequence reaction is used for production on an automatic sequencer. Visualization of chromatograms is carried out using the Benchling online algorithm (https://www.benchling.com/). Alleles are determined automatically using reference sequences using the BDRB32ex software script containing instructions in the Python programming language.

EFFECT: stable reproducibility of results when analyzing the number of DNA copies exceeding the sensitivity threshold of the test system.
