FIELD: medicine, clinical biochemistry.
SUBSTANCE: invention relates to a method for determination of nitrates and nitrites in lacrimal fluid. Method involves precipitation of proteins with 1.5 ml of 96% ethanol cooled to +4-8°C and 0.1 ml of distilled water in the final concentration 86.4% and then 2 ml of ammonium-chloride buffer at pH 9.6 ± 0.05 is added and volume is brought about to 10 ml. Then nitrates and nitrites are isolated through a cadmium column, color reaction is carried out with addition of N-naphthylethylenediamine dihydrochloride followed by photometry and determination of concentration of nitrates and nitrites by plot showing dependence of concentration from optical density. The concentration of nitrates and nitrites are calculated by the following formula: NOx (mcmole/l) = wherein C1 means concentration of nitrite ions calculated by calibration plot; NOx =
NO- 2/NO- 3. Invention allows assay of concentration of nitrates and nitrites in biological media of small volumes.
EFFECT: improved method of determination.
