FIELD: biotechnologies.
SUBSTANCE: method for production of heterogeneous biocatalyst for fermentative synthesis of aminobeta-lactam antibiotic based on intracellular hydrolase of alpha-aminoacid ethers from bacteria of Xanthomonas type, including damage of biomass in cells of Xanthomonas rubrilineans, extraction of ferment from it into water phase and its further immobilisation. Besides, damage of cells is carried out in stages, first damaging their integrity by cryogenic action, and then by treatment of water suspension of thawed cells by butyl acetate. Extraction of ferment into water phase is carried out in process of treated cell suspension heating first in conditions of spontaneously established gradient of pH, and then at constant value of pH. Produced cell-free extract is used for procedure of ferment immobilisation by means of its deposition by polyethylene glycol in the form of ferment aggregates and their further cross linkage in presence of glutaric dialdehyde. Thus obtained heterogeneous biocatalyst contains up to 0.95 g of protein/g of dry biocatalyst and has synthetase activity of up to 2200 ME/g of dry biocatalyst.
EFFECT: invention makes it possible to improve efficiency of biocatalyst production procedure, to increase activeness and stability of produced biocatalyst.
