FIELD: chemistry; biochemistry.

SUBSTANCE: invention relates to molecular genetics and cattle breeding. The method involves preparation of a nucleic acid sample, carrying out a polymerase chain reaction using two internal allele-specific primers and two external primers. The internal primers are characterised by absence in the 3rd position with 3' -ends of the nucleotide which is non-homologous to sequences of corresponding DNA chains of the kappa casein gene. Also the reverse external primer casR has specificity due to its annealing in exon 4 of the CSN3 gene. The DNA polymerase used is KlenTaq DNA polymerase, which enables amplification of A- and B-allele fragments of sizes 122 bps and 174 bps respectively, as well as amplification of the kappa casein gene with size of 250 bps, which enables control of efficiency of the reaction.

EFFECT: method increases specificity of PCR.
