METHOD OF OBTAINING VACCINE AGAINST ANIMAL PASTEURELLOSIS Russian patent published in 2010 - IPC A61K39/102

FIELD: medicine.

SUBSTANCE: invention relates to veterinary microbiology. Method includes cultivation of Pasteurella multocida of serological versions A, B, D and Pasteurella haemolytica in liquid nutrient medium. Each strain is inoculated separately in amount 10% to nutrient medium volume and cultivated at temperature 37°C to maximal level of accumulation of soluble superficial antigens in growth medium. Obtained bacterial mass is inactivated with formalin for 24 hours at temperature 37°C±1°C to final formalin concentration of 0.3%. After that inactivated bacterial mass of each strain is mixed in equal ratios, centrifuged at 6000-8000 rev/min, obtained precipitate is re-suspended in 0.1-0.2 M phosphate buffer with pH 7.2-7.4 to concentration 40-100 billion m.c./cm3 with further heating for 30-60 min at 56-60°C. Obtained mass is centrifuged at 10000-15000 rev/min, supernatant is separated and concentrated 3-5 times fold. Activity of superficial soluble antigens is determined in reaction of passive hemagglutination, and if activity of superficial soluble antigens of Pasteurella multocida of serological versions A, B, D and Pasteurella haemolytica is respectively 1:2048-4096, 1:4096-5120, 1:2048-8192 and 1:16-64, vaccine is obtained.

EFFECT: method allows to accelerate process of vaccine obtaining and increase its activity.
