SUBSTANCE: method for determining water soluble carbohydrates and starch from a single sample relates to agriculture and can be used in plant breeding for study of carbohydrate status of plant tissues and organs in ontogeny, but also in food and pharmaceutical industries to assess the quality of crops and medicinal raw materials, as it enables to determine simultaneously starch and soluble sugars. The proposed method for determining water soluble carbohydrates and starch from one sample of plant material is based on thermal extraction with an aqueous solution of a ground sample, followed by centrifugation, and acid hydrolysis of sludge containing starch. Determination of carbohydrates content is carried out in two solutions: in supernatant fluid - water-soluble carbohydrates, and in the solution after hydrolysis of sludge - starch, colorimetrically on intensity of colour change due to chemical interaction of carbohydrates with picric acid in alkaline medium with heating in a boiling water bath, and the calculation is carried out by calibration curve in comparison with the control solution.

EFFECT: this technique is simple, fast and reliable.
