FIELD: biotechnologies.

SUBSTANCE: this is a method of non-transgenic generation of a plant, resistant or tolerant to phosphor-methyl-glycine group herbicide. It includes introduction of recombinant oligonucleo base into plant cells with directed mutation in EPSPS gene for plant cells generation with mutant EPSPS gene expressing EPSPS protein which is mutant under one or several amino acid positions, where specified positions are chosen from the group, including Leu¹⁶⁰, Thr¹⁷⁹, Pro¹⁸³, Val¹¹⁴, Asp¹⁶⁴, Asn¹⁹³ and X¹⁹⁴ in EPSPS protein Arabidopsis (AF360224), or according to similar amino acid residue in EPSPS paralog, where X is not Leu; choice of a plant cell expressing enhanced tolerance to glyphosate, if compared to a corresponding wild plant cell; regeneration of a non-transgenic herbicide resistant or tolerant plant with mutant EPSPS gene from the specified plant cell. A plant generated under this method is provided.
