COMPOSITIONS AND METHODS INCLUDING VERSIONS OF CELLULASE WITH REDUCED AFFINITY TO NON-CELLULOSIC MATERIALS Russian patent published in 2013 - IPC C12N9/42 C12P19/02

Abstract RU 2486242 C2

FIELD: biotechnology.

SUBSTANCE: selected versions of cellulase are proposed, where the said versions are versions of the original cellulase, which amino acid sequence is identical to at least 75% amino acid sequence of SEQ ID NO: 3. At that one version comprises substitution at a position selected from the position 63, 77, 129, 147, 153, 157, 161, 194, 197, 203, 237, 239, 247, 254, 281, 285, 288, 289, 294, 327, 339, 344, 356, 378 or 382, and the second version of cellulase comprises additionally to one of the above mentioned substitutions a substitution at a position 146, 151, 189, 208, 211, 244, 277 or 405, where the positions are numbered according to the amino acid sequence of the reference cellobiohydrolase II (CBH2) presented in SEQ ID NO: 3, and where the substitution in one or more positions is the reason that the version of cellulase has a more negative net charge than the reference CBH2, and the version of cellulase has increased cellulolytic activity compared to wild-type cellulases. The methods of conversion of biomass and production of fuel with use of the said version of cellulose are disclosed.

EFFECT: invention enables to produce cellulases with increased cellulolytic activity compared to wild-type cellulases.
