FIELD: chemistry.
SUBSTANCE: invention relates to biochemistry. Described is single-chain antibody (scFv), binding with TNFα, containing domain VH and domain VL, bound with linker; wherein linker includes two cysteines, capable of forming intra-chain disulphide bond, and wherein amino acids between cysteines in linker sequence form loop fragment, when two cysteines are bound with each other by disulphide bond; and scFv has sequence SEQ ID No. 8. Described is application of amino acid linker for binding scFv VH and VL domains, wherein linker includes two cysteines, capable of forming intra-chain disulphide bond, and wherein amino acids between cysteines in linker sequence form loop fragment, when two cysteines are bound with each other by disulphide bond; with scFv having sequence SEQ ID No. 8.
EFFECT: invention improves pharmacokinetic properties (for instance half-life period in vivo, tissue permeability and time of residence time in tissue.
