FIELD: medicine.
SUBSTANCE: matrix of decellularised donor liver of mammal in amount of 1:1 for 8-12 hours at temperature of 4 °C is incubated in solution with pH of 7.4. Composition of solution: 138 mM NaCl, 2.67 mM KCl, 1.47 mM KH2PO4, 8.1 mM Na2HPO4, fibronectin and laminin at 10 mcg/ml, distilled water up to 1 l. One performs hepatocytes and bone marrow stem cells coculture for 2-4 days in amount of 2×106-15×106 per 1 cm of matrix. Matrix with cells volume of not less than 0.05 cm3 is mixed with collagen-containing hydrogel in amount of 3:1. Particle size of matrix from 200 to 700 mcm, not more than 50 mcm, porosity is 70-85 %. Obtained cell-engineering structure is implanted in liver parenchyma. Anticoagulants used in preventive dosage.
EFFECT: method enables improving effects of treating patient with hepatic failure due to activation of two-way interactions between implanted cell-engineering structure and recipient injured liver parenchyma.
