FIELD: immunology.
SUBSTANCE: invention relates to an immunological analysis, and concerns a sample analysis of blood or blood component for presence of free vitamin D, including vitamin D metabolites 25-hydroxyvitamin D or 1,25-dihydroxyvitamin D, where free vitamin D is circulating untied vitamin D fraction. Method includes steps of: (a) adding immobilised binding protein or antibody to 25-(OH)-vitamin D in sample; (b) mixing sample with diluent, said solvent contains 0.1% to 0.25% fluoroalkyl surfactant; (c) incubation of sample for period of time sufficient to provide binding of required amount of vitamin D with binding protein; (d) removing unbound whey and components of whey by flushing; (e) analysis of immobilized binding proteins or antibodies containing trapped vitamin D, connected with them by competitive binding using compound of vitamin D; (f) determination of concentration of labelled compounds of vitamin D associated with binding protein. Method also involves quantitative analysis of captured vitamin D via mass spectrometry.
