FIELD: chemistry.
SUBSTANCE: invention relates to a two-stage method of chromatography for purifying proteins, particularly monoclonal antibodies, using only four buffer solutions obtained from the mother solution. Method involves (a) first chromatographic stage including: passage of the said solution through the first chromatographic column; elution of crude protein from the first chromatographic column using the first elution buffer; and (b) second chromatographic stage including: passage of eluent of the crude protein obtained at the end of stage (a) through the second chromatographic column; extraction of a purified protein from the second chromatographic column using the second elution buffer, herewith each of the buffers contains Bis-Tris, acetic acid and water.
EFFECT: method enables fast production of batches of purified protein and reduction the time of using cleaning systems, that enables industrial purification of recombinant proteins.
