SUBSTANCE: methods of determining presence or absence of inserted nucleotide sequence in a certain site of insert in nucleic acid include: nucleic acid recovery from one sample or portions; bringing the nucleic acid into contact with a direct primer capable to bind to the nucleic acid above the site of insert; with the first reverse primer specific for the inserted nucleotide sequence, and with the second reverse primer capable to bind to the nucleic acid below the site of insert. Replicated nucleic acids are analyzed to determine presence or absence in the sample of an inserted nucleotide sequence.

EFFECT: use of the methods enables high-sensitive determination of zygosity in the analyzed sample.
