FIELD: biochemistry.

SUBSTANCE: present invention relates to biochemistry and immunology, in particular, to a dimeric antigen-binding molecule, which is crosslinked with antigen A and antigen B. Said molecule consists of two polypeptide chains, each of which comprises a variable light chain domain, specific to antigen A (VLA), variable heavy chain domain, specific to antigen B (VHB), variable light chain domain, specific to antigen B (VLB) and a variable heavy chain domain, specific to antigen A (VHA). Domains are arranged in following order VLA-VHB-VLB-VHA. VLA of first polypeptide chain is linked to VHA of second polypeptide chain to form an antigen-binding site for antigen A, VHB of first polypeptide chain is bonded to VLB of second polypeptide chain to form an antigen-binding site for antigen B, VLB of first polypeptide chain is linked to VHB of second polypeptide chain to form an antigen-binding site for antigen B and VHA of first polypeptide chain is linked to VLA of second polypeptide chain to form an antigen-binding site for antigen A. Present invention discloses a composition for treating diseases, in which antigens A and B are expressed and containing said antigen-binding molecule.

EFFECT: present invention due to such arrangement of variable domains in antigen-binding molecule VLA-VHB-VLB-VHA from N-terminal to C-terminal of polypeptide, makes it possible to crosslink between antigens A and B, improves efficiency of binding with target antigens.
