FIELD: biotechnology.
SUBSTANCE: proposed vaccine comprises recombinant pseudo-adenovirus particles on the basis of human adenovirus of fifth serotype bearing under the control of an inducible promoter, the full-length M2 protein gene and the full length protein gene NP, modified PEST motif and removing a nuclear localization signal, interconnected nucleotide sequence of FMDV 2A peptide. Vaccine is prepared by creating a consensus amino acid sequences of M2, and NP protein, NP protein sequence modifications PEST motif and removing a nuclear localization signal to obtain a genetic construct M2-2A-NP-PEST, the genes expressing these proteins under the control of a promoter inducible Tet-off. Preparation of plasmid pShuttle-tet-M2NP-PEST cloning M2-2A-NP-PEST genetic construct a shuttle plasmid carrying the regulatory elements Tet-off promoter to give plasmid pAd5-tet-M2NP-PEST homologous recombination using the plasmid pShuttle-tet-M2NP-PEST with a plasmid containing the genome of a recombinant human adenovirus of fifth serotype transfection plasmid pAd5-tet-M2NP-PEST in 293NEK cell multiplication, accumulation of particles of pseudo-adenovirus recombinant Ad5-tet-M2NP-PEST in the presence of doxycycline, the concentration and purification of these recombinant of pseudo-adenovirus particles using ultrafiltration and high performance liquid chromatography. Bringing pharmaceutically acceptable buffer concentration to 106-109 PFU/ml. The resulting preparation immunogenic and protective activity against influenza A virus of the following subtypes: H1N1, H2N3, H3N2, H5N2, H9N2.
EFFECT: invention can be used as a candidate universal vaccine against influenza A viruses of human and birds.
