SUBSTANCE: way of getting that includes cultivation of the polypeptide of Escherichia coli cells, containing nucleic acid encoding a polypeptide, regulation of pH values during cultivation using alkaline solution containing amino acid polypeptide selection of cells or cultural medium and, thus, obtaining a polypeptide. However, this concentration of amino acid in alkaline solution is higher than the solubility of amino acids in water at 20°C and at neutral pH. The concentration of amino acid in alkaline solution is 30 g/l or more. Alkaline solution represents a 10 % (weight/volume) or more of a solution of ammonia. Escherichia coli cell is a cell of auxotrophic on leucine and proline strains of Escherichia coli K12. Alkaline solution contains leucine and proline. Cultivating implement to achieve cell dry weight 20 g/l or more.
