FIELD: chemistry.
SUBSTANCE: disclosed is a method of producing a recombinant polypeptide. The method includes culturing CHO cells in a medium with the total amino acid content of about 40 to about 100 mM in conditions which include at least one temperature shift and at least one pH shift, and expression of the recombinant polypeptide. The cells are grown at a temperature of 33°C-38°C for at least 3 days, followed by changing the temperature to 30°C-37°C. The cells are grown at pH 6.8-7.5 for at least 2 days, followed by changing the pH to 6.0-7.1. The cells are kept at 30°C-37°C for at least 2 more days and at pH 6.0-7.1 for at least 1 day.
EFFECT: invention reduces cell death, increases the output of the end product and improves the quality of the end product.
