SUBSTANCE: invention relates to biotechnology, particularly to culturing cells and use thereof to produce polypeptides. A method of producing a recombinant post-translational modified polypeptide includes a production step. Recombinant CHO cells are cultured in a cell culture medium, the cell culture medium being a medium which is free of serum and protein, but contains 220-2500 mg/l choline chloride or an equimolar amount of another choline salt.
EFFECT: cell culture medium with high choline chloride content is particularly suitable for periodic cell culturing with injection, wherein cell vitality remains at a high level for a long period of time and with high polypeptide titres, although limited amounts of amino acids are used.
