METHOD OF POTATO MICROCLONING IN VITRO OF "ERMAK" POTATO VARIETY Russian patent published in 2017 - IPC A01H4/00

SUBSTANCE: method includes cultivating healthy potato plants in vitro by means of micro-grafting on a nutrient medium containing macro- and microelements according to the Murashige-Skoog formula, Fe-chelate, agar-agar, vitamins according to White's subscription, ascorbic acid and sucrose, obtaining regenerating plants and obtaining potato micro-tubers. Wherein etiolated seedlings (1-1.5 cm) are sterilized in the 0.1% diacid solution for 3-5 minutes, followed by a three-time wash with sterile distilled H2O. For cultivating micro-plants and forming micro-tubers, glass jars (900 ml) with a metal lid and an opening in the lid for introducing a nutrient media and micro-plants are used. The opening is closed with a cotton-gauze stopper and covered with foil. Sucrose of 81000-85000 mg/l, ferulic acid of 1-2 mg/l, kinetin of 1-2 mg/l, thiamine of 1-1.2 mg/l are introduced in the nutrient medium for tuberization at the temperature of 20°C. The daylight conditions are 8 hours (5000 lux), the conditions of total darkness - 16 hours, cultivation is carried out for 30 days before forming microtubers.

EFFECT: method allows to obtain healthy domestic potato free from latent virus infection and having excellent quality characteristics, is easy to use, environmentally safe, and can be used at potato growing enterprises.
