FIELD: biotechnology.
SUBSTANCE: invention relates to the field of biotechnology, namely to the technology of accelerated propagation of test-tube potato plants under in vitro conditions. The invention is a method for micropropagation of potatoes, including the propagation of test-tube potato plants in vitro using a Murashige and Skoog nutrient medium containing macrosalts, microsalts, Fe-chelate, agar-agar, White's vitamins and sucrose, glucose, agar. At the same time, preliminary recovery of the original potato plants from a viral infection is carried out by sterilizing the apical meristems (0.2-0.5 mm) in a 0.3% diacid solution for 3-5 minutes, followed by three washings with sterile distilled H2O. For the cultivation of healthy potato plants (explants), ferulic acid 0.5 mg/l, kinetin 1 mg/l, PPT 0.01 mg/l are added to the nutrient medium in the following ratio of components: macrosalts 50 mg/l, microsalts 1 mg/l, Fe2SO4·7H2O - 5.0 mg/l, CaCl2·2H2O - 440 mg/l, meso-inositol - 100 mg/l, thiamine - 1.0 mg/l, pyridoxine - 1.0 mg/l, sucrose - 20 000 mg/l, agar - 7000 mg/l, pH 5.7-5.8. Cultivation of plants (explants) is carried out in a culture room at illumination of 5000 lux, photoperiod of 16 hours, temperature of 25°C and air humidity of 70-80%.
EFFECT: method makes it possible to increase the efficiency of propagation of test-tube potato plants free from viral and bacterial infections under in vitro conditions, is easy to use and can be used at enterprises to obtain sterile seed potatoes.
