FIELD: biotechnology.

SUBSTANCE: method for DNA purification from paraffin blocks with a histological biomaterial is proposed. The paraffin is removed from the biomaterial sample, incubated and the biological material is precipitated by centrifugation. The precipitate is washed with 96% ethanol and treated with a denaturing buffer containing 3 M guanidine chloride, 10-20 mcl proteinase K solution at a concentration of 20 mg/ml and 10 mM Tris acetate. The resulting insoluble precipitate is precipitated by centrifugation. 1/3 volume of 96% ethanol is added to the supernatant and a DNA sample is applied to a column with fiberglass sorbent, for example, a "Biosilica" sorbent. The sorbent is washed with a buffer solution containing 3M guanidine chloride, 10 mM Tris-acetate pH 6.5, 25% ethanol, and then with a buffer solution containing 10 mM Tris-HCl pH 7.5, 0.1 M NaCl, 75% ethanol. The DNA is eluted with sterile distilled water.

EFFECT: invention allows to reduce the purification of DNA from paraffin blocks and increase the yield of DNA.
