SUBSTANCE: invention relates to biotechnology, in particular to a composition for inhibiting growth and stimulating apoptosis of cancer cells of colorectal cancer by blocking the function of the MCM4 and Livin genes. Specified composition comprises of a liposome for delivering the composition to target cells and a combination of two duplexes of oligonucleotide siRNA. First duplex is designed to block the genes of MCM4 and contains preparations of small interfering oligonucleotides 5'-C-U-C-A-Umet-C-U-C-U-Umet-A-C-C-C-A-C-A-G-G-dTs-dT-3' as a direct chain and 5'-C-C-U-G-U-G-G-G-Umet-A-A-G-A-G-A-Umet-G-A-G-dTs-dT-3' as an inverse chain. Second duplex is designed to block the genes of Livin and contains preparations of small interfering oligonucleotides 5'-G-G-A-A-G-A-G-A-C-U-Umet-U-G-U-C-C-A-Cmet-A-dTs-dT-3' as a direct chain and 5'-U-G-U-G-G-A-Cmet-A-A-A-G-U-C-Umet-C-U-U-C-C-dTs-dT-3' as an inverse chain. Umet - 2'-O-methyluridin-3'-phosphate; Cmet - 2'-O-methylcytidine-3'-phosphate; dTs - 2'-deoxythymidine-3'-phosphorothioate; dT - 2'-deoxythymidine-3'-phosphate.
EFFECT: invention makes it possible to increase the efficiency of inhibiting the growth and induction of apoptosis in malignant tumor cells, while reducing the side effects.
