FIELD: chemistry.

SUBSTANCE: method for amplifying and sequencing target loci in a nucleic acid sample is provided. Method comprises (a) bringing a nucleic acid sample containing the target loci into contact with a library of test primers containing at least 1,000 different test primers, wherein the concentration of each test primer is less than 20 nM; (b) amplifying the reaction mixture by polymerase chain reaction (PCR), wherein the PCR comprises an annealing step with a duration of more than 10 minutes; while simultaneously amplifying at least 1,000 different target loci and wherein (i) less than 20% of the amplified products are represented by dimers of the test primers, (ii) at least 80% of the amplified products are represented by target amplicons and (iii) at least 80% of the target loci are amplified; and (c) sequencing the amplified products.

EFFECT: invention improves efficiency of analysis while increasing its sensitivity and specificity.
