FIELD: biotechnology; medicine.

SUBSTANCE: invention relates to biotechnology and immunology. Method for the preparation of AmpT protein for simulating anti-cholera immunity in experimental animals is proposed. Method involves growing an avirulent strain of V. cholerae eltor 18950, washing the cells, followed by precipitation, destruction in an ultrasound disintegrant, treatment with DNA-ase and RNA-ase, and centrifugation. Supernatant is ultracentrifuged, the pellet of membrane proteins is dissolved in 1 % Triton X-100, ultracentrifuged. OmpT protein is isolated from the supernatant liquid previously pre-dialyzed against 20 mM Tris-HCl (pH 7.6), applying to the column with DE-52 cellulose. Proteins sorbed on the cellulose are eluted with a gradient of NaCl.

EFFECT: method provides an effective preparation for modeling immunity in experimental animals.
