SUBSTANCE: invention relates to the field of biology and experimental medicine. Method for evaluating pharmacological and toxic properties of substances – potential human AHR ligands – using Drosophila melanogaster flies is proposed. Drosophila melanogaster genome flies, transformed with UAS-AhR gene construction by hybridological synthesis, further include a tissue-specific gene construct, at the third stage of larval development are transferred to nutrient medium containing the test substance. UAS-AhR dpp-GAL4 flies which reach imago stage, the extremitas structures are assessed. UAS-AhR GMR-GAL4 flies eye structures are evaluated. UAS-AhR GMR-GAL4 and UAS-AhR elav-GAL4 flies, transcription level of AHR target genes are estimated. Obtained characteristics of the flies are compared and pharmacological and toxic properties of the test substance are determined by comparing the degree of morphological changes in extremitas structures and eye structures and transcription level of target human AHR genes of all flies groups.
EFFECT: invention provides fast, targeted and reliable selection of potential human AHR ligands.
