FIELD: biochemistry.
SUBSTANCE: invention relates to biochemistry, genetic engineering and biotechnology, in particular to a recombinant plasmid DNA pET31b-pHLIP. Said plasmid DNA encodes an amino acid sequence of a recombinant pH-dependent incorporating peptide and provides its synthesis in a fused protein with ketosteroid isomerase. Invention also relates to a method for obtaining said peptide. Present method involves the transformation of Escherichia coli cells of plasmid DNA pET31b-pHLIP, growing of transformed cells in a medium with ampicillin, induction of expression, subsequent cultivation, separation of cells by centrifugation, washing of inclusion bodies, their dissolution in a buffer with the addition of urea, purification by metal-chelate chromatography, subsequent separation of the peptide from ketosteroid isomerase and 6 histidine residues by using bromocyan.
EFFECT: present invention allows obtaining a recombinant pH-dependent incorporating peptide.
