FIELD: biotechnology, genetic and protein engineering. SUBSTANCE: single-stranded antibodies against tick-borne encephalitis virus are prepared by insertion of DNA fragment into plasmid vector pGEM1 encoding lider peptide of pectinase B (pel3) of Erwinia carotovora and genes of variable domains MKA E6B against tick-borne encephalitis virus in the composition of united DNA sequence. Constructed recombinant plasmid DNA pTLS provides expression of single-stranded antibodies against tick-borne encephalitis virus in cells of bacterium E. coli. Obtained chimeric antibodies retain the conformation of variable domain of the parent MKA and are able to interact with domain E2 of surface glycoprotein of tick-borne encephalitis virus. Invention proposes the recombinant strain of bacterium E. coli B-716 as producer of single-stranded antibodies against tick-borne encephalitis virus that can be used for development of medicinal agents of the novel generation against tick-borne encephalitis virus.
