FIELD: chemistry.
SUBSTANCE: invention relates to a new method for producing 2,4-di-C1-6alkyl pyrimidine-5-ol of formula (I), in particular, 2,4-dimethylpyrimidine-5-ol, and to a new starting product, a nitrophenyl ester of general formula (V). Method also includes the steps of preparing a compound of formula (V). Most preferred compound of formula (I), obtained according to the proposed method – 2,4-dimethylpyrimidine-5-ol, is a key intermediate compound for obtaining a compound that has an antagonistic effect on an orexin receptor and is suitable as a treatment for insomnia. Method for obtaining the compounds of formula (I) or its salt, (I) (V) where R1 and R2 represent C1-6alkyl groups that are the same or different from each other include the step of hydrolyzing a compound of formula (V) or a salt thereof. New compound of formula (V) or its salt is obtained by reacting a compound of formula (IV) or its salt with the compound of formula (VI) or its salt (IV) (VI). Compound of formula (IV) used or its salt is obtained by reacting a compound of formula (III) with N,N-dimethylformamide dimethyl acetal, (III). Compound of formula (III) is obtained by the reaction of 4-nitrophenol with a compound of formula (VII) , and X represents chlorine or bromine, and R1 and R2 at all stages have the above values. Preferably at all stages of the process as R1, and R2 represent methyl groups. Compound of formula (I) is obtained in high yield (93 %). New compound (V) used in the method is an inexpensive and readily available starting material and allows to conduct the process regioselectively.
EFFECT: production of the product and impurities is easily controlled and does not require the use of reagents and intermediate compounds that cause health problems.
