FIELD: nanobiotechnology.
SUBSTANCE: invention relates to physiology and nanobiotechnology of plants. Method involves growing plants in the presence of heavy metals of copper and nickel and subsequent evaluation of resistance. Growth of plants in presence of heavy metal nanoparticles is carried out for 48 hours at concentration of 0.125–1 M copper nanoparticles, or copper oxide, or nickel. Then roots of wheat germs are homogenised with subsequent DNA extraction and their separation by method of horizontal gel electrophoresis. DNA damage coefficient Kdam is determined by fluorescence intensity of high-molecular DNA Khm fragments 30,000–3,000 bp to low-molecular DNA Klm fragments <3,000 bp. At Kdam ≤0.4 – high-stable wheat, at Kdam = 0.4–0.59 – stable, with Kdam = 0.6–0.79 – medium-stable, Kdam = 0.8–1.09 – unstable, and at Kdam ≥1.1 – absolutely unstable wheat.
EFFECT: method enables assessing differential rapid diagnostics of resistance of wheat at the stage of seedlings to heavy metal nanoparticles.
