FIELD: biotechnology.
SUBSTANCE: invention relates to the field of biotechnology and plant breeding, namely to a method for simultaneous sequencing of the nucleotide sequences of a panel of genes underlying the traits of durum wheat important for the breeding process. This method includes the following steps: isolation of a high-molecular fraction of DNA from 4-day durum wheat seedlings (HMW DNA), enrichment of genomic DNA with high-molecular fragments; designing gRNAs specific to significant durum wheat genes and in vitro synthesis of gRNAs on a double-stranded DNA matrix molecule, assembly of Cas9/gRNA ribonucleoprotein complexes, Cas9-targeted nanopore sequencing, bioinformatic analysis of the data obtained: assembly and alignment on the genome.
EFFECT: expansion of the range of solutions for wheat gene sequencing.
