PREPARATION BASED ON LYTIC MYCOBACTERIOPHAGE AND A METHOD FOR PRODUCTION THEREOF Russian patent published in 2019 - IPC C12N15/00

Abstract RU 2691439 C1

FIELD: biotechnology.

SUBSTANCE: invention relates to a preparation for inactivation (lysis) of intracellular tuberculosis mycobacteria based on a lytic mycobacteriophagous strain which inactivates intracellular tuberculosis mycobacteria, which is a mycobacteriophage D29 included in a bilayer phospholipid liposome of 0.4 mcm, and a method for production thereof, comprising separating and purifying phage particles from bacterial lysates of M.smegmatis with subsequent inclusion into phospholipid liposomes, where separation and purification of phage particles from bacterial lysates M.smegmatis is carried out by ion-exchange chromatography on a Q-sepharose column, obtained purified mycobacteriophage in concentration of not less than 108 pfu/ml is shaken with phospholipid film for 5–10 minutes, obtained particles are extruded through filter with 5 micron pores and subsequent 20-fold extrusion using filters with pore size of 0.4 mcm, concentration and separation of a fraction of mycobacteriophage included into the liposome by centrifugation at 13±2 thousand rpm for 30±5 minutes, using a phospholipid film obtained from phosphatidylcholine, cholesterol and tween-80, taken in a weight ratio of 5±1:1±0.8:1.2±0.2.

EFFECT: what is presented is a preparation for intracellular mycobacterium tuberculosis inactivation (lysis).
