FIELD: biotechnology.
SUBSTANCE: disclosed is a set of targeting structures comprising a first construct comprising a first homology region with a target genome of the microorganism, an endonuclease recognition site and a first portion of the selectable marker, and a second construct comprising a second portion of the selectable marker, an endonuclease recognition site and a second homology region with the target microorganism target genome, wherein the first portion fragment of selectable marker is overlapped with the second portion fragment of selectable marker, enabling recombination between the first and second portions of the selectable marker. Invention also relates to a method of modifying a genome of a microorganism and a method of producing a microorganism.
EFFECT: present invention provides effective targeting and modification of microorganism target gene.
