FIELD: biology.
SUBSTANCE: invention relates to genetics and molecular biology and concerns a method for obtaining translocants between any loci belonging to different chromosomes in cells of budding yeast Saccharomyces cerevisiae. Disclosed is a method of producing site-directed translocations in eucaryotic cells of budding yeast Saccharomyces cerevisiae, consisting in lithium-acetate transformation of ura3Δ0/ura3Δ0 cells by a translocation cassette carrying a URA3 gene flanked by homology regions of more than 30 base pairs to two different chromosomes. Homology regions define a cartridge integration point. Correctness of the translocation is checked during the PCR reaction with the primers flanking the regions of homology of the used cartridge, and sequencing the obtained PCR product.
EFFECT: disclosed is a method for producing site-directed translocations in Saccharomyces cerevisiae cells.
