FIELD: medicine.
SUBSTANCE: invention refers to medicine, namely to oncology, and can be used for evaluation of hydrogen peroxide content in tumor cells. That is ensured by examining the effect on tumor cells of an antitumor preparation represented by genetically encoded HyPer protein. Antineoplastic preparation is a photosensitizer. After photosensitizer is accumulated by cells, photodynamic reaction is initiated by irradiation of cells with laser with wave length corresponding to spectral range of photosensitizer absorption. Fluorescence is recorded in range of 500–560 nm. Intracellular content of hydrogen peroxide is sequentially measured at wavelengths of 405 nm and 488 nm by laser scanning microscopy using a fluorescent sensor. Thereafter, the value of the ratio between the intensity of the fluorescence signal when excited at wavelength of 488 nm is calculated to the intensity of the fluorescence signal when excited at wavelength of 405 nm (I488/I405), on which intracellular content of hydrogen peroxide is determined.
EFFECT: invention enables determination of the place and time of formation of hydrogen peroxide in the irradiated tumor cell, as well as detection of dose dependence.
