FIELD: measurement.
SUBSTANCE: group of inventions relates to measurement equipment. Method for calibrating an apparatus which includes an optical system capable of imaging fluorescent radiation from a plurality of reaction areas, such as wells of a sample holder, includes: performing calibration of the range of interest (RI) to determine positions of reaction sections on the image, wherein calibration of the RI reduces errors in determining reaction areas by minimizing at least one factor from the following group: dye saturation in multiple reaction sections, grid rotation, gain variation and optical radial distortion; performing calibration of the pure dye to determine the contribution of the fluorescent dye used in each reaction section by comparing the initial spectrum of the fluorescent dye with the calibration data of the pure spectrum of the fluorescent dye, wherein the reaction sections contain identical dye concentrations to provide pure spectral values for each well of the sample holder, and the sample holder is a calibration plate with a staggered configuration; calibrating the device normalization to determine the normalization coefficient for the filter; and RNase P validation is carried out to validate the instrument's ability to distinguish two different amounts of one sample.
EFFECT: technical result consists in ensuring resistance to acceptance of suboptimal calibrations, improved objectivity and uniformity during such an activity.
