FIELD: chemistry.
SUBSTANCE: invention refers to analytical chemistry, namely to methods for quantitative determination of toxic components in implants based on polylactide-glycolide (PLGA) by gas chromatography. Method for simultaneous determination in one sample of quantitative determination of toxic components (acetone, ethyl acetate, lactide) in PLGA implants, including scaffolds for culturing cells based on polylactide-glycolide PLGA and calcium phosphate FC (hydroxyapatite Ca10(PO4)6(OH)2 and β-tricalcium phosphate Ca3(PO4)2), by gas chromatography on capillary column based on 100 % polyethylene glycol involves filtration of sample solution of scaffold in chloroform using syringe filter with pore size of 0.45 mcm, chromatography of obtained solution using flame-ionisation detector with temperature of evaporator and detector 250 °C and temperature mode of column: plateau 80 °C – 2 min, heating 10 °C/min to 170 °C – plateau 1 min, heating 20 °C/min to 220 °C – plateau 17.5 min; pressure of carrier gas (nitrogen) 40 kPa, flow division 1:20, 0.5 mcl of obtained solution is introduced by micro-syringe in the evaporator, calibration dependences for acetone (in range of 8·10-4–0.2 %) are plotted, lactide (in range of 6·10-3–0.25 %), ethyl acetate (in range of 1·10-4–0.075 %), according to which content of toxic components in the sample is calculated.
EFFECT: technical result is possibility of simultaneous in one sample of quantitative determination of acetone, ethyl acetate, lactide in implants and scaffolds.
