SUBSTANCE: strain of hybrid cultured animal cells Mus musculus 1E10, producer of mouse monoclonal antibody 1E10, which specifically binds to the protective antigen Bacillus anthracis in Domain IV region, is proposed. The specified strain is deposited in the “State Collection of Pathogenic Microorganisms-Obolensk” Federal Budgetary Institution of Science “State Scientific Center of Applied Microbiology and Biotechnology” with the number N-89. Mouse monoclonal antibody 1E10, neutralizing the lethal toxin Bacillus anthracis, specifically binding to the protective antigen Bacillus anthracis in the region of Domain IV, is proposed. 1E10 antibody is characterized by equilibrium dissociation constant 9.156410-10 M and is represented by two identical heavy and two identical light chains forming the IgG1 molecule, with amino acid sequences of light chain SEQ ID NO: 1 and Fd of heavy chain SEQ ID NO: 2 fragment. Also proposed is a chimeric recombinant monoclonal antibody xi1E10, which neutralizes the lethal toxin Bacillus anthracis, obtained by replacing the constant domains of the light chain and heavy chain of the above 1E10 antibody with similar regions of human immunoglobulin G1, expressed in eukaryotic cells of ExpiCHO-S line, specifically binding to the Bacillus anthracis protective antigen in Domain IV region. xi1E1 antibody is characterized by equilibrium dissociation constant 1.266310-9 M and is represented by two identical heavy and two identical light chains with amino acid sequences of light chain SEQ ID NO: 3 and heavy chain SEQ ID NO: 4.

EFFECT: group of inventions ensures preparation of antibodies with a high affinity and highly specific binding to the Bacillus anthracis protective antigen in the region of Domain IV, which has a neutralizing effect against the Bacillus anthracis lethal toxin.
