SUBSTANCE: group of inventions relates to chemistry of antisense compounds, molecular biology and medicine. Disclosed are compounds capable of reducing expression of RNA or protein 4R of isoform Tau, based compositions and methods for modulating Tau mRNA splicing in an animal. In one version, the disclosed compound comprises a modified oligonucleotide consisting of 15–30 linked nucleosides and having a nitrogen base sequence comprising a complementarity region, which contains at least 15 adjacent nitrogen bases, complementary parts of equal length of nitrogen bases 121708–122044 of SEQ ID NO: 1, wherein modified oligonucleotide contains 2'-substituted sugar fragment. In another version, the target complementarity region lies within nitrogen bases 121873–121898 of SEQ ID NO: 1.
EFFECT: such compounds and methods are suitable for treating, preventing or alleviating neurodegenerative diseases associated with Tau in an individual in need thereof.
