SUBSTANCE: invention relates to biotechnology and is a VHH domain, a humanized VHH domain, or an camelized VH domain for use in a pharmaceutical composition for the treatment of diseases or disorders in humans, the blood of which contains preexisting antibodies that can bind to the exposed C-terminal region of the VH domain even then when the VH domain contains a C-terminal extension in which the amino acid residue at Position 89 is L and the amino acid residue at Position 11 is V, wherein the amino acid positions are indicated using Kabat numbering, with the VHH domain, humanized VHH domain, or camel VH domain having reduced binding to preexisting antibodies, present in blood or serum samples from subjects suffering from diseases or disorders, compared to a VHH domain, humanized VHH domain or camelized VH domain, where the amino acid residue at Position 89 is V, and the amino acid residue at Position 11 is L.
EFFECT: invention makes it possible to obtain improved variable domains of the immunoglobulin heavy chain, which, when they have an exposed C-terminal site or end, will be less likely to bind to pre-existing antibodies/factors such as those found in blood or serum samples obtained from testees.
