FIELD: biotechnology; pharmacology.
SUBSTANCE: invention relates to biotechnology and pharmacology and is a conjugate of recombinant human granulocyte-macrophage colony-stimulating factor (GM-CSF) with alendronic acid, a representative of bisphosphonates, having an increased affinity for bone marrow target cells. A targeted colony-stimulating agent is characterized by the fact that it is a conjugate of the protein of granulocyte-macrophage colony-stimulating factor GM-CSF with alendronic acid, linked to each other at the carboxy group of the protein in the form of a monoadduct having high affinity for hydroxylapatite, obtained by the reaction in the presence of a water-soluble linking agent 1-ethyl-3-[3-dimethylaminopropyl] carbodiimide (EDC) and having the following structure:
where ALN is alendronic acid; GM-CSF is a granulocyte-macrophage colony-stimulating factor. The conjugate of granulocyte-macrophage colony-stimulating factor with alendronic acid was obtained at their stoichiometric ratio per 1 mol of GM-CSF - 1 mol of alendronic acid.
EFFECT: creation of a hemo-stimulating targeted agent based on a conjugate of GM-CSF and alendronic acid with the properties of a vector molecule directed to target cells of the bone marrow.
