SUBSTANCE: invention relates to biotechnology. Described is a nucleic acid vector for producing a replication-defective retroviral vector particle containing a non-mammalian animal origin of replication and capable of storing at least 25 thousand base pairs (tbp) of DNA, while the vector is selected from: bacterial artificial chromosome; yeast artificial chromosome; artificial chromosome derived from P1; phosmids; or cosmids, characterized in that the specified nucleic acid vector contains sequences of the nucleic acid of the retrovirus encoding: gag and pol proteins and env protein, where each sequence of the nucleic acid of the retrovirus has its own promoter in the nucleic acid vector. Also disclosed is a method for producing replication-defective retroviral vector particle.

EFFECT: invention expands the arsenal of means for gene therapy.
